Audax Italiano's traditional rivals are Palestino, a team founded by the Palestinian colony residing in Chile, and Unión Española, which originates from the Spanish community present in the country. Each match between them is called the Clásico de colonias, however, the match against Unión Española is the one with the longest tradition. The origin of the rivalry with Unión is found in the fact that both institutions are representatives of two large foreign colonies in the country, in addition to the good presentations of both teams in the 1920s —when the rivalry began to take shape—, which made that a large number of spectators went to witness this match which not infrequently, especially during the 1930s, ended in clashes involving fans of both squads.
Starting in the late 1920s, Audax also maintained a rivalry with Colo-Colo, whose roots lay in the good performance of both teams during that period. The rivalry continued until the 1950s, a period in which the match was called the "Creole Classic", involving the only two clubs that had a fully national squad.
